L’obiettivo di questa ricerca era valutare i range di adiponectina (ACRP-30) e leptina nel siero e nel liquido sinoviale (SF) di donne obese con gonartrosi maggiore (GOA) e ricercare le loro correlazioni con la posizione scientifica e l’entità dell’OA biochimica biomarcatori: metalloproteinasi di matrice (MMP) 1, MMP-9 e inibitore tissutale della metalloproteinasi (TIMP-1). Il gruppo studiato era composto da 39 donne obese che hanno subito un intervento di artroplastica completa del ginocchio a causa di GOA.
Lo stadio di GOA è stato valutato in risposta ai punteggi Knee Society Rating (KSS), Ahlbäck e Kellgren-Lawrence (Ok-L). Erano stati prelevati sangue periferico e SF. Gli intervalli di biomarcatori scelti sono stati valutati utilizzando kit ELISA . Lo stadio implicato di ACRP-30 nel siero valutato era di 8393,80 ng/mL e in SF, 774,33 ng/mL, il focus principale di leptina nel siero era 32.040,74 pg/mL e in SF, 27.332,12 pg/mL. Intervalli di leptina nel siero e SF correlati con l’indice di massa corporea (BMI), ( p = 0,0005 e p = 0,0002, rispettivamente).
Gli intervalli di ACRP-30 nel siero sono stati correlati con i punteggi scientifici (Ahlbäck: p = 0,0214; Ok-L: p = 0,0146). ACRP-30 in SF correlato con ACRP-30 nel siero ( p = 0,0003), tendeva a correlare negativamente con MMP-1 nel siero ( p = 0,0598) e correlare positivamente con pro-MMP-1 in SF ( p = 0,0600). Per concludere, questa ricerca conferma le correlazioni tra le concentrazioni di ciascuno, leptina e ACRP-30, valutando siero e SF. Nelle femmine obese, i livelli di leptina migliorano con l’IMC e lo stadio sierico ACRP-30 migliora con ulteriori livelli superiori di GOA. Infine, la leptina gli intervalli erano stati correlati con il focus sierico di TIMP-1, uno dei tanti marcatori biochimici di GOA.
Le cellule adipose pancreatiche delle persone con diabete di tipo 2 mostrano un ridotto esercizio adipogenico e lipolitico
I problemi di peso, in particolare l’accumulo di grassi viscerali, aumenteranno la possibilità di diabete di tipo 2 (T2D). Lo scopo di questa ricerca era di studiare l’influenza del diabete di tipo 2 sul deposito di grassi pancreatici. Sono stati raccolti cuscinetti di grasso pancreatico da 17 pazienti parzialmente pancreatectomizzati (PPP), preadipociti pancreatici isolati e differenziati in vitro. I malati sono stati raggruppati utilizzando HbA1c in normali tolleranti al glucosio (NGT), prediabetici (PD) e T2D. I profili del trascrittoma di preadipociti e adipociti sono stati valutati mediante RNAseq.
La sensibilità all’insulina è stata stimata quantificando la fosforilazione di AKT su western blot. La capacità lipogenica è stata valutata con Otaining rosa olio, esercizio lipolitico attraverso il lancio di acidi grassi. Gli elementi secreti sono stati misurati utilizzando ELISA . La valutazione comparativa del trascrittoma di preadipociti e adipociti indica una sovraregolazione difettosa dei geni che governano l’adipogenesi (NR1H3), la lipogenesi (FASN, SCD, ELOVL6, FADS1) e la lipolisi (LIPE) durante la differenziazione delle cellule da T2D-PPP.
Inoltre, il rapporto tra leptina /adiponectina mRNA era maggiore in T2D che in NGT-PPP. I preadipociti e gli adipociti di NGT-PPP erano più sensibili all’insulina rispetto alle cellule T2D-PPP per quanto riguarda la fosforilazione di AKT. L’accumulo di trigliceridi era paragonabile negli adipociti NGT e T2D. Indipendentemente da un’eccessiva espressione dei recettori NPR1 e NPR2 negli adipociti NGT e T2D, la lipolisi è stata stimolata da ANP 1,74 volte esclusivamente nelle cellule NGT.
Questa stimolazione è stata ulteriormente elevata dal dipiridamolo inibitore della PDE5 (3,09 volte). Il dipiridamolo e la forskolina hanno aumentato il recettore della lipolisi in modo indipendente di 1,88 volte e 1,48 volte, rispettivamente, esclusivamente nelle cellule NGT. In conclusione, lo stato metabolico influenza in modo persistente la differenziazione e la lipolisi degli adipociti pancreatici. Queste alterazioni potrebbero peggiorare l’evento di T2D.

I problemi di peso sono legati al pericolo eccessivo e alla prognosi infausta della maggior parte dei tumori al seno (BC). I problemi di peso favoriscono la proliferazione delle cellule BC modulando la produzione di adipochine, insieme ad adiponectina (anti-neoplasticadipochina), leptina (adipochina cancerogena) e mediatori dell’infiammazione.
Nell’ambito della ricerca attuale abbiamo studiato i risultati anti-proliferativi di liraglutide (LG; farmaco antidiabetico e per la riduzione del peso) su cellule BC umane MCF-7 coltivate in terreno condizionato da cellule staminali derivate da tessuto adiposo in sovrappeso (ADSCs-CM) e se o meno questo impatto è mediato dalla modulazione delle adipochine nelle ADSC e nella maggior parte delle cellule tumorali. La proliferazione è stata studiata utilizzando il controllo di vitalità AlamarBlue, il test di formazione di colonie e la valutazione del ciclo cellulare.
Gli intervalli e l’espressione delle adipochine e dei loro recettori sono stati saggiati utilizzando ELISA e RT-PCR. LG ha provocato l’inibizione del 48% della proliferazione MCF-7 in ADSCs-CM in sovrappeso, ha ridotto la formazione di colonie e ha indotto l’arresto della parte G0/G1. LG ha inoltre ridotto i gradi dei mediatori dell’infiammazione, soppresso l’espressione della leptina , mentre elevati intervalli di mRNA di adiponectina e dei loro recettori nelle ADSC in sovrappeso e nella maggior parte delle cellule tumorali coltivate in ADCS-CM in sovrappeso.
In conclusione, LG potrebbe mitigare lo sviluppo delle cellule BC in argomenti in sovrappeso; per questo motivo può benissimo essere utilizzato per la prevenzione scientifica e/o il rimedio del BC in soggetti in sovrappeso. Potrebbe aiutare a migliorare i risultati del trattamento e ridurre il tasso di mortalità nei pazienti in sovrappeso con BC.
Aptasensor per la quantificazione della leptina tramite PCR Amplificazione di brevi DNA-aptameri
La quantificazione delle proteine è storicamente effettuata mediante test di immunoassorbimento enzimatico ( ELISA ), che comporta lunghi tempi di preparazione. Per battere questo, i nuovi approcci utilizzano gli aptameri come scelta alternativa agli anticorpi. In questo documento, presentiamo un metodo nuovo di zecca per quantificare le proteine con brevi aptameri di DNA mediante risposta a catena della polimerasi (PCR) che porta a istanze di protocollo più brevi con limiti di rilevamento relativamente migliorati.
La metodologia proposta presenta una nuova opzione per quantificare contemporaneamente sia la proteina obiettivo che i corrispondenti brevi aptameri di DNA, che ci consente inoltre di caratterizzare completamente l’efficienza degli aptasensori. La leptina umana viene utilizzata come proteina obiettivo per convalidare questo sistema, poiché è considerato un biomarcatore vitale per la ricerca sull’obesità.
Nei nostri esperimenti, abbiamo raggiunto il limite inferiore di rilevamento di 100 pg/mL in meno di 2 ore, un limite influenzato dalla dissociazione fissata dall’aptamero leptinico, che può benissimo essere migliorato scegliendo un aptamero in più. Grazie al metodo semplice ed economico, questo sistema potrebbe essere impiegato per implementazioni Lab-On-Chip e per una rapida quantificazione “in loco” delle proteine.
La leptina stimola Akt nella maggior parte delle cellule tumorali esofagee attraverso una serie di piccole GTPasi sensibili all’atorvastatina
I problemi di peso sono un problema pericoloso per l’esofago di Barrett e l’adenocarcinoma esofageo. Il tessuto adiposo secerne l’ormone leptina . La leptina è un problema di sviluppo per un certo numero di tipi di cellule, insieme alle cellule di Barrett e alle cellule di adenocarcinoma esofageo. Le statine sono correlate alla riduzione delle cariche dell’esofago di Barrett e della maggior parte dei tumori esofagei e mostrano risultati antitumorali in vitro. I meccanismi di tali risultati non saranno del tutto stabiliti.
Abbiamo ora esaminato le conseguenze della leptina e della statina liposolubile, atorvastatina, sulla segnalazione tramite proteine leganti GTP monomeriche e Akt. La proliferazione e l’apoptosi sono state valutate nelle cellule OE33. L’esercizio di Akt è stato quantificato mediante ELISA cellulare e test della chinasi in vitro. Particolari inibitori di piccole molecole e un assemblaggio dominante-negativo erano stati usati per ridurre l’esercizio di Akt. Piccole GTPasi erano state inibite utilizzando la trasfezione di plasmidi dominanti negativi, inibitori della prenilazione e pretrattamento con atorvastatina.
La leptina ha stimolato l’esercizio di Akt e la proliferazione cellulare e ha inibito l’apoptosi indotta dalla camptotecina in un metodo Akt-sensibile. La leptina ha indotto la fosforilazione di Dangerous e FOXO1 in un metodo Akt-sensibile. La leptina ha attivato Ras, Rac, RhoA e cdc42. La trasfezione di plasmidi dominanti-negativi ha confermato che l’ attivazione di Akt indotta dalla leptina richiedeva Ras, RhoA cdc42 ma non Rac. L’atorvastatina ha inibito l’ attivazione indotta dalla leptina di Ras, RhoA, cdc42 e Akt. Il co-trattamento con mevalonato ha impedito questi risultati dell’atorvastatina.
